چکیده

The Agilent 1260 Infi nity Hybrid SFC/UHPLC System offers a fast, reliable, green, robust, and cost-effective solution by coupling the advantages of HPLC and the benefi ts of supercritical fl uid chromatography (SFC). This Application Note describes the conversion of the United States Pharmacopeia (USP) normal-phase HPLC method for cholecalciferol assay to an SFC method using serially coupled columns. The 1260 Infi nity Hybrid SFC/UHPLC System was used to perform both the HPLC as well as the SFC methods. The results show that the SFC method meets the system suitability criteria, is 2x faster, and achieves 26x lower solvent costs compared to the USP method. Robustness tests on the SFC method demonstrated excellent robustness for routine analysis. Authors Vinayak AK Syed Salman Lateef Agilent Technologies, Inc. Bangalore, India mAU

Agilent Application Solution Transfer of a USP method for tolazamide from normal phase HPLC to SFC using the Agilent 1260 Infi nity Hybrid SFC/UHPLC System

Normal phase liquid chromatography methods often have long run times and involve environmentally toxic/costly solvents. Supercritical fl uid chromatography (SFC) methods on the other hand, are faster, inexpensive, and eco-friendly. SFC involves the use of low viscosity supercritical carbon dioxide, which can be operated at fl ow rates up to three times higher than liquid chromatography (LC) wit...
